- 12/23 Rule: Recombination occurs only between a 12bp spacer RSS and a 23bp spacer RSS. Prevents direct V-to-J heavy chain joining.
- RSS (Recombination Signal Sequence): Conserved heptamer (CACAGTG), nonamer (ACAAAAACC), and spacer. Guides RAG1/RAG2 complexes.
- P-nucleotides: Templated palindromic sequences added to coding ends when RAG cleaves hairpin loops asymmetrically.
- N-nucleotides: Non-templated nucleotides added randomly by Terminal deoxynucleotidyl Transferase (TdT).
- Productive Joint: Junction maintaining correct open reading frame (multiple of 3 bp) with no premature stop codon.
